Hi John the amplifier is a weak spot on the Lexus Vehicles - members have reported faults on the LS430 and LS460 before. Sadly I have had this problem - it turned out to be the power supply on the amplifier - I had mine repared by SVT services in Glasgow for £285 inc return postage.

When mine packed up I heard a loud crackle and thats it - all the functions on the screen still worked inc volume counting up but no sound at all.

On the 430 there is afuse for the amp - might be worth looking if this is the case with yours and checking it.

There is a simple diagnostic you can do through the touchscreen for the LS430 - not sure if it will work on your vehicle.

thought to delve deeper as i was removing rear trim anyway

simple task to pull out the lot located AMP in rear boot space

awkward to access unless you break it down patiently

took me an hour fortunately i have the space and tools

although i am NOT a mechanic .

passenger side fuse not blown need to check others

Blimey you've been busy and well done to you stripping all that down. If the controls are working then I'd think the amp is at fault. If possible try a different amp just to test. Even a 2 channel amp will test if sound comes out.
