Has anyone managed to send a planned route to their car (2019 NX 300h) Navigation from "My Lexus".

Not recently, but I did manage it when I first got the car and was playing around. That route still shows in my 'favourites.' 

From memory I had to turn the wi-fi on, using my mobile telephone hotspot. I'll have another go later on and see whether I can still do it!

Yes - same model as yours but in grey!  I set my car to auto connect to my home wifi, you have to tell My Lexus to Share to Car.  

Make your route in 'JOURNEY PLANNER' the select 'Share to Car' or 'Download to USB'.

I’ve now tried all this, car links to my home WiFi but no planned route “appears” in the Nav menu.

Has the Route been sent to the car in your Journey Planner ?

If so it Downloads when you next start your car with your phone's personal Hot Spot set ON

Drops into Favourites from memory.

I find it for some reason only works when I first go to POS, before it did it automatically

I've tried all the solutions offered and the route still disappears into the ether, I guess it's a glitch that isn't worth fixing since apple Car Play will be retrofitted later in the year.
