I need to replace the pieces of trim down the sides of the windscreen (on the outside) as the paint is peeling off of the pieces oftrim currently in place.

I have purchased the two pieces from Lexus but have no idea on how to fit them.

Does anyone know if the windscreen has to come out for the trim to be replaced?

looking at the manual for the 430 it says, "use a trim removing tool to remove the trim" very helpfull, it looks like it is held on by a series of friction clips. At a guess tape around the old one then try to pull/prise it off straight up away from the car, if not then try asking a windscreen company, they must remove them all the time, shouldn't think it would be much of a job when you've got the tool and a bit of experience.
