Just about all modern cars have electronic speedos driven off a squarewave pulse from the gearbox. Toyota's standard colour for the pulse wire to the back of the speedo is purple/white (but I don't know for sure if this is the case on the IS) - if it's there scope it and see if you have a pulse. You might be able to pursuade someone in a dealership to tell you what colour the wire is, or even fault find it.

Another possibility (what I did) is to call up a Satnav company, such as VDO Dayton, and ask the tech support there - they keep lists of speedo pulse connections for their fitters

Hi,

On some RWD cars the speedo sensor reads and the teeth/cogs that are on the drive shaft from the diff, maybe  the sensor might just need cleaning...

Just cover all bases. Another possible location for your speedo might be on the wheel bearing at the back wheels in which the ABS sensor picks up the rotation of the wheel. Don't know if that's the case with Lexus as it was the case in my last car and nobody knew it except for the dealer. Probably wrong but it's another option.
