I seem to recall a similar post some time ago and it turned out to be a seized lock mechanism.

You could try fully lowering the window and with the help of a torch have a look inside the door panel to see if there is a way of getting it open or get a local auto locksmith to do the job.

Once opened you can then remove the door trim and see what is not working in there.

potentially linked issue.

Sometimes my driver door self locking mechanism makes an awful screeching noise like it's crying out for oil or grease. Took it to Lexus Teesside and couldn't demonstrate it for them. Then I realised it was on the second press of the handle tit - ie the double locking mechanism. They checked it today and found 3 of the 4 doors do it. They will correct it but have to strip the doors down - in the meantime I'm going to make sure I don't double lock the doors or they might seize like lexusmadman's
